A French study published in the British Medical Journal has warned the public about the potential link between the prolonged use of Pfizer-made hormonal drugs for birth control and menopausal relief and an increased risk of intracranial meningiomas in women.
Intracranial meningiomas are mostly non-cancerous brain tumors that slowly grow in the layers of tissue covering the brain and spinal cord. However, due to the pressure they exert on the brain, they usually lead to complications, including 40 percent of central nervous system cancers. Surgical removal is often necessary, but it usually damages nearby brain structures.
The study, which analyzed data from the French national healthcare system, involved 18,061 women who underwent surgery for intracranial meningioma from 2009 to 2018. Each case was compared with five women without the tumor. Researchers tracked the usage of various progestogen drugs, including hydroxyprogesterone, dydrogesterone, medrogestone, medroxyprogesterone acetate, promegestone, dienogest and levonorgestrel.
According to the study, led by Noémie Roland, a general practitioner and epidemiologist at the French National Health Insurance, the use of Depo-Provera (medroxyprogesterone acetate), a popular contraceptive injection, along with the menopausal hormone therapy drugs medrogestone and promegestone for more than a year increased the risk of intracranial meningiomas in women. Depo-Provera increased the risk of intracranial meningioma by 5.6 times, while medrogestone and promegestone were associated with a 4.1- and 2.7-fold increased risk, respectively.
Meaning, about four in every one thousand 30-year-old women would develop intracranial meningiomas at the age of 80. The use of Depo-Provera for more than a year increased the risk by 5.6 times. Moreover, the figure could still rise to 20 in every one thousand.
“In countries for which the use of medroxyprogesterone acetate for birth control is frequent [74 million users worldwide], the number of attributable meningiomas may be potentially high,” Roland concluded.
The researchers have also warned the public that their findings could not draw a direct cause-and-effect link between medroxyprogesterone acetate, medrgoestone and promegestone and intracranial meningiomas because their study is observational. Instead, they urged their colleagues to conduct more research regarding the findings.
“Women taking the commonly used birth control pills or hormone replacement therapy are not at increased risk of meningioma,” said Pharoah. “It is important that women do not stop using their birth control pills without consulting their doctor. The risk of meningioma is extremely small. If I were currently using Depo-Provera because of the advantages of a long-acting injectable contraceptive I would continue to do so. In short, current or previous users of Depo-Provera do not need to worry about their very small risk of meningioma.”
“We are aware of this potential risk associated with long-term use of progestogens and, in collaboration with regulatory agencies, are in the process of updating product labels and patient information leaflets with appropriate wording,” Pfizer said.
